Memantine (1-amino-3,5-dimethyladamantane) (Fig. 1) is a moderate affinity, uncompetitive NMDA receptor antagonist and is approved for the symptomatological treatment of moderate to severe Alzheimer s disease (2002 in EU and in the 2003 USA in 2003).
Merz (Germany) as the patent holder markets memantine under the trade name Axura in several countries and the licensing partners Forest Labs Inc and Lundbeck Pharmaceutical use the trade names Namenda in the USA and Ebixa respectively. Memantine blocks NMDA receptor with an average affinity of c.a. 1 M (varying upon receptor subtype, type of cells used and readout), by binding to a site within the channel pore (Fig. 2). As NMDA receptors activation is required for many forms of learning of memory (Morris et al., 1986; Collingridge and Bliss, 1995) this mechanism of action (MoA) might seem difficult to comprehend.
